New Cholesteryl Ester Transfer Protein from Indonesian Herbal Plants as Candidate Treatment of Cardiovascular Disease

نویسندگان

چکیده

BACKGROUND: There is a strong negative relationship between high-density lipoprotein cholesterol (HDL-C) and the risk of cardiovascular disease (CVD). Cholesterol ester transfer protein (CETP) glycoprotein transporter that transfers esters to very low-density (LDL-C). The CETP inhibitor new strategy against CVD because its ability increase HDL-C. Various Indonesian plants have not been optimally used, in silico phytochemical screening these showing potential as inhibitors still limited. AIM: This study for exploring phytochemicals treatments. METHODS: We screened 457 registered herbal database met Lipinski’s rule five. Their molecular structures were downloaded from PubChem database. three-dimensional dalcetrapib (the standard) obtained data bank (http://www.rcsb.org/pdb/) with 4EWS code ZINC ZINC03976476 code, respectively. CETP–dalcetrapib binding complexes validated 5 times using AutoDock Vina 1.1.2 software. Interactions molecularly docked same software visualized Pymol 1.8× RESULTS: Dalcetrapib had docking score −9.22 kcal/mol bound at Ser230 His232 residues. 11 lower scores than dalcetrapib, but only L-(+)-tartaric acid, chitranone, oxoxylopine could interact residue. These are commonly found Tamarindus indica, Plumbago zeylanica, Annona reticulata, CONCLUSION: L-(+)-Tartaric show silico.

Plasma cholesteryl ester transfer protein.

The plasma lipoproteins are continuously remodeled during their transit through the plasma compartment, owing to the action of lipid metabolizing enzymes and lipid transfer processes. These activities have a major effect on the composition, size, and concentration of the lipoproteins. Lipid transfer processes involving neutral lipids and phospholipids are mediated by specialized plasma proteins...

Association of Circulating Cholesteryl Ester Transfer Protein Activity With Incidence of Cardiovascular Disease in the Community

Background—Plasma high-density lipoprotein cholesterol concentration is related inversely to the risk of cardiovascular disease (CVD). Inhibiting cholesteryl ester transfer protein (CETP) activity raises high-density lipoprotein cholesterol and may be cardioprotective, but an initial clinical trial with a CETP inhibitor was stopped prematurely because of increased CVD in treated patients, raisi...

The yin and yang of cholesteryl ester transfer protein in cardiovascular disease.

Cholesteryl ester transfer protein (CETP) is a 79-kDa glycoprotein, present in humans, rabbits, primates, and hamsters but absent in rodents, dogs, horses, cows, and pigs, that facilitates transfer of cholesteryl ester from high-density lipoprotein (HDL) particles to low-density lipoprotein (LDL)–very-low-density lipoprotein (vLDL) particles in exchange for triglycerides, thereby participating ...

Cholesteryl ester transfer-protein modulator and inhibitors and their potential for the treatment of cardiovascular diseases

Elevated low-density lipoprotein (LDL) cholesterol and lowered high-density lipoprotein (HDL) cholesterol are important risk factors for cardiovascular disease. Accordingly, raising HDL cholesterol induced by cholesteryl ester transfer protein (CETP) inhibition is an attractive approach for reducing the residual risk of cardiovascular events that persist in many patients receiving low-density L...

Targeting cholesteryl ester transfer protein for the prevention and management of cardiovascular disease.

Epidemiologic studies have shown that the concentration of high-density lipoprotein cholesterol (HDL-C) is a strong, independent, inverse predictor of coronary heart disease risk. This identifies HDL-C as a potential therapeutic target. Compared with low-density lipoprotein cholesterol (LDL-C)-lowering agents, however, currently available HDL-raising drugs are relatively ineffective. Consequent...
